Traditional PFAS analysis by mass spectrometry (MS) is time-consuming, as laborious sample preparation (e.g., extraction and desalting) is necessary. Herein, we report fast detection of PFAS by paper spray (PS)-based MS techniques, which employs a triangular-shaped filter paper for sample loading and ionization (≤ 3 min per sample). In this study, PS-MS was first used for direct PFAS analysis of drinking water, tap water, and wastewater. Interestingly, food package paper materials can be directly cut and examined with PS-MS for possible PFAS contamination. For samples containing salt matrices which would suppress PFAS ion signal, desalting paper spray mass spectrometry (DPS-MS), was shown to be capable of rapidly desalting, ionizing and detecting PFAS species such as per-fluorooctanoic acid (PFOA) and per-fluorosulphonic acid (PFOS). The retention of PFAS on paper substrate while salts being washed away by water is likely due to hydrophilic interaction between the PFAS polar head (e.g., carboxylic acid, sulfonic acid) with the polar filter paper cellulose surface. The DPS-MS method is highly sensitive (limits of detection:1.2-4.5 ppt) and can be applicable for directly analyzing soil extract and soil samples. These results suggest the high potential of PS-MS and the related DPS-MS technique in real-world environmental analysis of PFAS.

A fluorescence biosensor was developed for the ultrasensitive detection of the available lead in soil samples by coupling with DNAzyme and hairpin DNA cyclic assembly. The biorecognition between lead and 8-17 DNAzyme will cleave the substrate strands (DNA2) and release the trigger DNA (T), which can be used to initiate the DNA assembly reactions among the hairpins (H1, H2, and H3). The formed Y-shaped sensing scaffold (H1-H2-H3) contains active Mg2+-DNAyzmes at three directions. In the presence of Mg2+, the BHQ and FAM modified H4 will be cleaved by the Mg2+-DNAyzme to generate a high fluorescence signal for lead monitoring. The linear range of the fluorescence biosensor is from 1 pM to 100 nM and the detection limit is 0.2 pM. The biosensor also exhibited high selectivity and the nontarget competing heavy metals did not interfere with the detection results. Compare with the traditional method (DTPA+ICP-MS) for the available lead detection, the relative error (Re) is in the range from -8.3 % to 9.5 %. The results indicated that our constructed fluorescence biosensor is robust, accurate, and reliable, and can be applied directly to the detection of the available lead in soil samples without complex extraction steps.

The development of effective strategies for the detection of oxytetracycline (OTC) in soil is of great importance for preserving agri-environmental safety and human health. Herein, a novel photoactive material of metal-organic framework (MOF) derived ZnO nanopolyhedra/graphitic carbon nitride (ZnO/g-C3N4) heterojunction was designed by mixing calcination of zeolite imidazole framework-8 (ZIF-8) and melamine. A self-powered photoelectrochemical aptasensor for the sensitive and selective detection of OTC in soil was proposed using ZnO/g-C3N4 as the photoanode. The photoactivity of the MOF derived ZnO nanopolyhedra was regulated effectively by the introduction of g-C3N4, which resulted in a 7-fold increase in the photocurrent of the ZnO nanopolyhedra at a bias potential of 0 V. It was assigned to the higher carrier density of ZnO/g-C3N4. By virtue of the amplified photocurrent of ZnO/g-C3N4, the specificity of the OTC aptamer and the anti-interference ability of the self-powered sensing method, the designed aptasensor demonstrated the advantages of a wide linear range (0.005-200 nM), low limit of detection (1.49 × 10-3 nM), good selectivity and good reproducibility. For real soil sample analysis, satisfactory recoveries were obtained and further verified by ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS).

Abstract Many soil microorganisms' i.e., bacteria and fungi produce secondary metabolites called antibiotics. These are used for the treatment of some of the bacterial, fungal and protozoal diseases of humans. There is a need for isolation of a broad spectrum of antibiotics from microorganisms due to the emergence of antibiotic resistance. In the present study two antibiotic producing bacteria Klebsiella pneumoniae and Bacillus cereus were isolated from pharmaceutical and poultry feed industry of Hattar, Haripur Pakistan. Total 10 waste samples were collected from different industries (Marble, Ghee, Soap, Mineral, Steel, Poultry Feed, Pharmaceutical, Qarshi, Cosmetic and Glass). Thirty-three bacterial strains were isolated from industrial wastes of these ten different industries. Fourteen out of thirty-three bacterial strains exhibited antimicrobial activities against at least one of the test microbes considered in this study including Escherchia coli, Staphylococcus aureus and Salmonella typhi. The bacteria were isolated by standard serial dilution spread plate technique. Morphological characterization of the isolates was done by Gram staining. Nine bacterial isolates out of fourteen were initially identified as B. cereus and five as K. pneumoniae through biochemical characterization. The antibacterial activities were tested by well diffusion method. Maximum number of antibiotic producing bacteria were isolated from pharmaceutical and poultry feed industry based on the results of primary screening, the most potential isolates S9, S19, S20, S22 and S23 were selected for secondary screening. The maximum activity against E. coli and S. aureus was recorded by bacterial isolate S19 i.e zones of inhibition of 6.5mm and 9mm while S20 showed 7.5mm and 6mm zones respectively. Molecular identification was carried out on the basis of 16S rRNA sequence analysis. Finally, the isolates were identified as B. cereus accession number LC538271and K. pneumoniae accession number MT078679. Analysis of bacterial extract S20 through GC-MS indicated the presence of 8 compounds of diverse nature and structure. Present study suggests that wastes of pharmaceutical and poultry feed industry may have antibiotic producing bacteria. These bacteria could be utilized for the production of antibiotics. B. cereus and K. pneumoniae isolated from wastes of poultry feed and pharmaceutical industries have the potential to produce antibiotics and could be used to control the microbial growth.

As bactérias foram isoladas pela técnica de placa de diluição em série padrão. A caracterização morfológica dos isolados foi feita por coloração de gram. Nove isolados bacterianos de 14 foram inicialmente identificados como B. cereus e cinco como K. pneumoniae por meio de caracterização bioquímica. As atividades antibacterianas foram testadas pelo método de difusão em poço. O número máximo de bactérias produtoras de antibióticos foi isolado da indústria farmacêutica e de ração avícola com base nos resultados da triagem primária, os isolados mais potenciais S9, S19, S20, S22 e S23 foram selecionados para a triagem secundária. A atividade máxima contra E. coli e S. aureus foi registrada pelo isolado bacteriano S19, ou seja, zonas de inibição de 6,5 mm e 9 mm, enquanto S20 mostrou zonas de 7,5 mm e 6 mm, respectivamente. A identificação molecular foi realizada com base na análise da sequência 16S rRNA. Finalmente, os isolados foram identificados como B. cereus número de acesso LC538271 e K. pneumoniae número de acesso MT078679. A análise do extrato bacteriano S20 por meio de GC-MS indicou a presença de oito compostos de natureza e estrutura diversas. O presente estudo sugere que resíduos da indústria farmacêutica e de ração para aves podem conter bactérias produtoras de antibióticos. Essas bactérias podem ser utilizadas para a produção de antibióticos B. cereus e K. pneumoniae isolados de resíduos de rações de aves e indústrias farmacêuticas têm potencial para produzir antibióticos e podem ser usados ​​para controlar o crescimento microbiano.

A gram-stain-negative, endo-spore forming, facultatively anaerobic, motile, rod-shaped bacterial strain SM69T, isolated from soil samples of Rohtak, Haryana, India was characterized using polyphasic approach. White colonies were 2-3 mm, in diameter and growth occurred between 20 and 55 °C, pH 6.0-10.0 with 0-2.0% (w/v) NaCl. Based on 16S rRNA gene sequence similarity the strain is placed in the genus Paenibacillus as it is closely related to 'Paenibacillus tyrfis MSt1T' (99.7%) and P. elgii SD17T (99.6%). The cell wall peptidoglycan contained meso-diaminopimelic acid. The dominant fatty acids included anteiso-C15: 0 (50%), C16: 0 (12%) and anteiso-C17: 0 (10%). Major polar lipids were diphosphatidylglycerol, phosphatidylethanolamine and phosphatidylglycerol. The size of the draft genome was 7,848,017 bp, with 53.1% G+C content. dDDH (51.6%) and ANI (93.5%) of strain SM69T with its close relatives indicates that it represents a novel species, for which the name Paenibacillus oleatilyticus sp. nov. (Type strain SM69T = MCC 3064T = JCM 33981T = KACC 21649T) is proposed.

Although triple-quadrupole inductively coupled plasma-mass spectrometry (ICP-MS/MS) has become an attractive technique for the measurement of long-lived radionuclides, the abundance sensitivity, isobaric and polyatomic ions interferences seriously restrict the application. The spectral peak tailing and uranium hydrides (UH+, UH2+) from 238U have a serious influence on the accurate measurement of 239Pu and 240Pu, especially for the ultra-trace level plutonium isotopes in the higher uranium sample. A new method was developed using ICP-MS/MS measurement in mass-shift mode with collision-reaction gas combined with a chemical separation procedure. As O2 readily converted Pu+ ion to PuO2+, while disassociated the interfering diatomic ions of interfering elements (U, Pb, Hg, Tl, etc.), the interferences from these elements were completely eliminated if plutonium was detected as PuO2+ at the m/z more than 270. By the mass filter in MS/MS mode combined with O2 as reaction gas the lower peak tailing of 238U+ (<5 × 10-12) was significantly suppressed. By this way, the 238UO2H+/238UO2+ atomic ratio was reduced to 4.82 × 10-9, which is significantly lower than that of other collision-reaction gas modes. Interferences from Pb, Hg and Tl polyatomic ions were also completely eliminated. Thus, accurate measurement of ultra-trace level 239Pu in high uranium sample solutions with the 239Pu/238U concentration ratio of 10-10 was achieved by the mass-shift mode with 0.15 mL/min O2/He + 12.0 mL/min He as collision-reaction gas, and high elimination efficiency of uranium interferences up to 1014 can be obtained by combination with the chemical separation using a single UTEVA resin column. The developed method can be applied to accurately determine the fg level 239Pu in high uranium samples, such as large-size deep seawater, deep soil and sediment, uranium debris of nuclear fuel.

@#Soil sample is one of the important evidence that can be found in crime scene. Unknown soil sample can be analysed and compared with reference sample in order to determine the origin as its physical and chemical components possess unique characteristics. The purpose of this study is to determine the physical and chemical characteristics of soil from oil palm plantations in Perak, Malaysia to assist forensic investigation. Total of 97 topsoil samples were collected from three different oil palm plantations in Perak. Particle size distribution was obtained using dry sieving technique and colour of soil sample was examined under three conditions that are dry, moist and ashed. Soil pH was measured using pH meter and percentage of composition of soil organic matter (SOM) was determined by weighing the sample before and after ignition. Result showed that the composition of particle size <0.18mm was within the range of 5.57-21.11% whereas for particle size between 0.18mm - 0.6mm was within 31.62 - 52.96% and 25.78-66.86% for particle size >0.6mm. The color mode of soil after oven dried, moistened and ashed was greyish brown (10YR 5/2), very dark greyish brown (10YR 3/2) and light yellowish brown (10YR 6/4) respectively. Soil pH was in the range of 5.79 – 6.70. The percentage of SOM was between 3.29 - 20.48%. The physical and chemical characteristics of soil analysed in this study from three different locations of oil palm plantations varied and it is possible to discriminate these locations based on the analysis highlighted in this study.

A novel actinomycete strain, designated CFH S0054T, was isolated from a soil sample collected from Xiaokongshan in Tengchong, Yunnan Province, in south-west PR China. Phylogenetic analysis based on its 16S rRNA gene sequence indicated that strain CFH S0054T belonged to the genus Nocardia. Based on the results of 16S rRNA gene sequence analysis, strain CFH S0054T was most closely related to Nocardia concava IFM 0354T (98.3 %), Nocardia inohanensis NBRC 100128T (97.9 %), Nocardia otitidiscaviarum NBRC 14405T (97.9 %) and Nocardia heshunensis CFH S0067T (97.8 %). Strain CFH S0054T contained C16 : 0, C18 : 1 ω9c, C18 : 0 10-methyl (TBSA) and summed feature 3 as major cellular fatty acids and MK-8 (H4ω-cycl) as the major respiratory quinone. The polar lipid profile included phosphatidylethanolamine, diphosphatidylglycerol, phosphatidylinositol, phosphatidylinositol mannoside and unknown phospholipids. The cell wall contained meso-diaminopimelic acid, and the whole-cell hydrolysates were arabinose, galactose, glucose, fructose, ribose and mannose. The genome of strain CFH S0054T was 7.9 Mbp with a G+C content of 68.4 mol%, the average nucleotide identity (ANI) values between strain CFH S0054T and N. concava IFM0354T were low (ANIm<87.7 % and ANIb=84.1 %). On the basis of phylogenetic, physiological and chemotaxonomic data, strain CFHS0054T is considered to represent a novel species of the genus Nocardia, for which the name Nocardiayunnanensis sp. nov. is proposed, The type strain is CFH S0054T (DSM 46763T=JCM 30082T).

A dually responsive fluorescent probe for determination of U(IV) and mercury(II) ions was synthesized. The probe consists of a cytosine-rich hairpin DNA loaded with silver nanoclusters (DNA-AgNCs). The fluorescence of the AgNCs is found to be quenched by UO2(II) at pH 5.0 and Hg(II) at pH 7.0 due to combined static and dynamic quenching. Under the optimal conditions, the green fluorescence of the DNA-AgNCs, best measured at excitation/emission wavelengths of 420/525 nm, decreases in the 4.0 to 75 pM UO2(II) concentration range, and in the 0.3 to 8.0 nM Hg(II) concentration range. The respective detection limits are as low as 1.8 pM and 0.1 nM. The method was successfully applied to the determination of UO2(II) and Hg(II) in (spiked) pond and taps waters and in soil extracts. Graphical abstract A label-free DNA was designed to synthesize green-fluorescent silver nanoclusters (AgNCs) and used for rapid dual detection of uranyl ions (UO2(II)) at pH 5.0 and of mercury ions (Hg(II)) at pH 7.0 in environmental samples.

The total accumulative stockpiles of gangue from long-term coal mining exceed 1 billion tons and occupy 182 square kilometers, and 50 million tons of additional gangue are generated per year in Shanxi, a major energy province in China. The objective of this study was to examine whether exposure to village soils affected by gangue stacking would disrupt thyroid hormone system homeostasis and eventually affect endocrine system and development, using zebrafish (Danio rerio) as a model organism. The zebrafish embryos were exposed to village soil leachates at 0, 1:9, 1:3 and 1:1 from 1 to 120 h postfertilization (hpf), and the sample caused a dose-dependent increase in the mortality and malformation rate, and decrease in the heart rate, hatching rate and body length of zebrafish larvae. Importantly, the soil leachate alleviated the whole-body triiodothyronine (T3) and thyroxine (T4) levels at higher concentrations, and altered the expression of the hypothalamic-pituitary-thyroid (HPT) axis-regulating genes crh, trh, tshß, nis, tg, nkx2.1, pax8, hhex, ttr, dio1, dio2, ugt1ab, trα, and trß and the PAH exposure-related genes ahr2 and cyp1a. These findings highlight the potential risk of thyroid hormone disruption and developmental toxicity from soil samples around coal gangue stacking areas.

Sampling and handling of soils and rhizosphere soil are very critical steps for obtaining representative microbial cultures and genomic material from these environments. Attention to position in the landscape of a sampling site, previous management of the site, time of year, and depth of sampling is important to assure representative samples. Detailed protocols are provided to assist environmental microbiologists and molecular biologists in proper sampling and handling of soils that serve as the source of cultures and DNA for subsequent use in important experiments carried out in the laboratory.

In this work, we proposed the functionalization of a surface plasmon-polariton (SPP)-supported gold grating surface with the metal-organic framework (MOF-5) for sensitive, selective and reproducible surface-enhanced Raman scattering (SERS) detection of organophosphorus pesticides. Homogeneous distribution of plasmon intensity along the Au grating surface ensures the high reproducibility of SERS results (deviation of Raman peak intensity does not exceed the 4% along the sample). The surface-assisted growth of thin MOF-5 film was accomplished in two steps procedure: (i) covalent grafting by 4-carboxyphenyl groups and (ii) the immersion of samples in the mother liquid of MOF-5. Proposed SERS chip proved itself to be a perfect analytical probe for the detection of organophosphorus pesticides with high reliability and low detection limit up to 10-12 M. Moreover, selective detection and recognition of several relevant organic contaminants (azo-dye, mycotoxin, and pesticide) from the simulated soil was successfully demonstrated. All SERS measurements were performed using portable Raman spectrometer and can easily be expanded to environmental conditions. Our work combines the high affinity of organic contaminants to the MOF-5 with excellent plasmonic excitation on the surface plasmon-polariton supported structure and shows the way to the realization of closed-to-ideal analytical SERS chip.

A novel solid phase microextraction fiber named graphene oxide incorporated poly acrylamide-ethylene glycol dimethacrylate (GO-poly AM-EDGMA) monolithic fiber has been successfully prepared in a fused silica capillary tube (250 µm, i.d.) via thermally initiated polymerization using acrylamide (AM) as the monomer, ethylene glycol dimethacrylate (EGDMA) as the crosslinker, dimethylformamide dispersed by GO as porogens, and then obtained by removing 1 cm wall from one end of the fused silica capillary tube. The fiber has large surface area (536 m2 g-1), pore capacity (0.694 cm3 g-1) good thermal stability (up to 320 ℃), long service life and good reproducibility (RSD <5% throughout 110 times) which facilitated for high throughput headspace solid phase microextraction (HS-SPME) coupled to gas chromatograph (GC) analysis. The HS-SPME/GC method using the new fiber was evaluated by the determination of five organophosphate esters (OPEs) in soil samples coupled to flame photometric detector (FPD). The proposed HS-SPME-GC/FPD method yielded satisfactory limits of quantification (0.03 ng g-1∼0.24 ng g-1); linearity (≥0.99), good intra- and inter-day precision expressed as relative standard deviations for a single fiber were in the range of 5.2-9.0% and 4.8-9.0%, respectively, and fiber-to-fiber reproducibility was in the range of 5.9-9.7%. The method was applied for the analysis of OPEs in environmental soil samples and the relative recoveries were found to be in the range from 80.1 to 105.6%. Based on these features, the new fiber has great potential for widespread use as a high throughput trace analysis tool.

A Gram-stain-positive, aerobic, motile with peritrichous flagella, rod-shaped bacterium, designated CFH S0170T, was isolated from a soil sample collected from Catba island in Ha Long Bay, Hai Phong City, Vietnam. Comparison of the 16S rRNA gene sequences showed that strain CFH S0170T belonged to the genus Paenibacillus and showed closest relationship with Paenibacillus vulneris CCUG 53270T (98.1 % similarity). Phylogenetic analysis demonstrated that the novel candidate formed a coherent branch with P. vulneris CCUG 53270T and Paenibacillus yunnanensis YN2T. Furthermore, the novel strain shared 87.2 % rpoB gene sequence similarity with P. vulneris CCUG 53270T. Growth of strain CFH S0170T occurred at 10-40 °C, pH 6.0-8.0 and with 0-2.0 % (w/v) NaCl. Strain CFH S0170T contained mannose, glucose and rhamnose as the major whole-cell sugars. The cell-wall peptidoglycan contained meso-diaminopimelic acid, glutamic acid, lysine and aspartic acid. The polar lipid profile contained diphosphatidylglycerol, phosphatidylethanolamine, glycolipids and phospholipids. The dominant cellular fatty acids included anteiso-C15 : 0 and C15 : 0. The genomic DNA G+C content was 50.9 mol%. On the basis of phenotypic, chemotaxonomic and phylogenetic analysis, strain CFH S0170T is affiliated to the genus Paenibacillus, but could be distinguished from other valid species of this genus. It is concluded that strain CFH S0170T should be considered to represent a novel species of the genus Paenibacillus, for which the name Paenibacillus esterisolvens sp. nov. is proposed. The type strain is CFH S0170T (=KCTC 33624T=BCRC 80802T).

A Gram-stain negative, aerobic, motile, non-spore-forming and rod-shaped bacterial strain, designated YIM 730227T, was isolated from a soil sample, collected from Karak district, Khyber-Pakhtun-Khwa, Pakistan. The bacterium was characterized using a polyphasic taxonomic approach. Pairwise comparison of the 16S rRNA gene sequences showed that strain YIM 730227T is closely related to Phenylobacterium lituiforme FaiI3T (97.5% sequence similarity), Phenylobacterium muchangponense A8T (97.4%), Phenylobacterium panacis DCY109T (97.1%), Phenylobacterium immobile ET (97.1%) and Phenylobacterium composti 4T-6T (97.0%), while also sharing 98.0% sequence similarity with Phenylobacterium hankyongense HKS-05T after NCBI blast, showing it represents a member of the family Caulobacteraceae. The major respiratory quinone was Q-10 and the major fatty acids were C16:0, summed feature 8 (comprising C18:1ω7c and/or C18:1ω6c), C18:1ω7c 11-methyl and C17:0. The polar lipids were phosphatidylglycerol, unidentified glycolipids, phospholipid and unidentified lipid. The G + C content of the genomic DNA was 68.2 mol%. The DNA-DNA relatedness values of strain YIM 730227T with P. hankyongense HKS-05T, P. lituiforme FaiI3T, P. muchangponense A8T, P. panacis DCY109T, P. immobile ET and P. composti 4T-6T were 31.3 ± 0.6, 26.1 ± 0.2, 24.3 ± 0.1, 21.8 ± 0.9, 19.8 ± 0.6 and 18.2 ± 1.1%, respectively, values lower than 70%. Besides the morphological and chemotaxonomic characteristics, phylogenetic analyses of 16S rRNA gene sequences and the biochemical characteristics indicated that the strain YIM 730227T represents a novel member of the genus Phenylobacterium, for which the name Phenylobacterium terrae sp. nov. (type strain YIM 730227T = KCTC62324T = CGMCC 1.16326T) is proposed.

The relative discrepancies between the gamma spectrometry efficiency of RGU, RGTh, RGK reference materials and some soil samples have been studied using a MCNP model of a real HPGe detector. It has been shown that, in a specified geometry, efficiencies differences depend on the sample elemental composition. The elemental compositions of RGU-1, RGTh-1 reference materials and a soil sample have been evaluated using X-Ray fluorescence (XRF) method and used in the MCNP simulation along with RGK-1 and six other soil samples with different elemental compositions to calculate their efficiencies in different gamma ray energies. To estimate the maximum relative efficiencies differences between soil samples and reference materials, five soil samples with higher attenuation properties were selected from a large data set of soils elemental compositions. The results show that the efficiency differences between soil samples and reference materials are almost ignorable for more than 100 KeV gamma energies. It strongly depends on the sample attenuation factor in the lower energies, so use of a self-attenuation correction is essential for radionuclide counting in low energies gamma rays. Results show about 8 percent discrepancy between RGU and two soil samples efficiencies in 63.2 KeV energy.

The aim of this study was to investigate the soil microbial communities of a phosphogypsum waste heap. The soil microbial community structures can differ over time, as they are affected by the changing environmental conditions caused by a long-term exposure to different kinds of pollutions, like is the case of soil in the post-production waste area in Wislinka (in the northern part of Poland) currently undergoing restoration. Our analyses indicated that the most abundant phyla were Proteobacteria, Acidobacteria, and Actinobacteria, and generally such an abundance is common for most of the studied soils. The most dominant class were Alphaproteobacteria, with their participation in 33.46% of the total reads. Among this class, the most numbered order was Sphingomonadales, whereas among this order the Sphingomonadaceae family was the most abundant one. The Sphingomonadaceae family is currently in the center of interest of many researchers, due to the ability of some of its members to utilize a wide range of naturally occurring organic compounds and many types of environmental contaminants. This kind of knowledge about microbial populations can support efforts in bioremediation and can improve monitoring changes in the contaminated environments.

RESUMO

Stigmatomma is the most speciose ant genus in the subfamily Amblyoponinae. In the present paper, the worker caste of a new species is described, S. luyiaesp. n., which was collected from a soil sample in a subtropical evergreen broad-leaved forest in Taiwan. An identification key to the females of Stigmatomma species with 11 antennomeres occurring in Asia is also provided.
